Back in the laboratory, I identified the species of the collected flies under a microscope as usual, determining whether each individual was Drosophila melanogaster, and established isofemale lines. Because it was still mid-summer and very hot, I was not able to collect many flies, but I still managed to establish several isofemale lines. Using these isofemale lines derived from the newly collected natural population, I performed allele-specific PCR to examine whether resistance-associated point mutations were present in actual field populations at three amino acid sites thought to contribute particularly to organophosphate resistance.

実験室に戻り、お決まりのようにキイロショウジョウバエの種の同定を行い、1雌由来系統を作製しました。真夏の、まだまだ暑い盛りだったので、あまりショウジョウバエを採集できませんでしたが、それでも数系統1雌由来系統を作製することができました。これらの、新たに採集した自然集団に由来する1雌由来系統を使って、実際に対立遺伝子特異的PCRを行い、有機リン剤抵抗性に特に寄与しているとされる3つのアミノ酸サイトにおいて、抵抗性型の点突然変異が、実際の野外集団に存在しているか否か検討を加えてみました。
